It'd be cool if one could just send an email to the sender of the email through the web interface. We already have the anonymous messaging system in place, at least in opencore, so it'd be nice to extend that. (if this issue should go somewhere in open core, ie it's not listen specific enough, let me know). If I see an email to a list from an interesting person, but I don't want to necessarily jump on the thread, then I can just send a private message to them.
So the interface would have 'reply list' and 'reply sender' options.

This is already there, at least to an extent. Mails in the archive that are from members of the site (who are signed up to the list as members of the site) have their email addresses replaced by a link to their author page, from which you should be able to send them a message without ever having known their email address. Of course this is not a reply to the message, and I can see the utility of a reply-off-list feature, though I'm not sure I'd put a high priority on it.
